Semax Research Overview

Introduction

Semax is a synthetic heptapeptide derived from adrenocorticotropic hormone (ACTH). Developed in Russia in the 1980s, it has been studied for its effects on cognitive function, memory, and neuroprotection in animal and in vitro models.

Mechanism of Action

ACTH Fragment Activity

Semax corresponds to amino acids 4-10 of ACTH. Unlike full-length ACTH, Semax lacks corticosteroid-stimulating activity but retains neurotrophic and cognitive effects observed in animal studies.

BDNF Expression

Research indicates Semax may upregulate brain-derived neurotrophic factor (BDNF) expression in the hippocampus and frontal cortex in animal models.

Serotonergic and Dopaminergic Systems

Animal studies suggest Semax may influence serotonin and dopamine neurotransmitter systems, though the precise mechanisms remain under investigation.

Current Research

Cognitive Research

Animal studies have investigated Semax's effects on learning, memory consolidation, and attention using various behavioural paradigms.

Neuroprotection Research

In vitro and animal studies have explored Semax's potential neuroprotective effects in models of oxidative stress, hypoxia, and excitotoxicity.

Pain Research

Some research has investigated Semax's effects on pain perception and analgesia in animal models.

Chemical Properties

  • **Sequence:** Met-Glu-His-Phe-Pro-Gly-Pro (7 amino acids)
  • **Molecular formula:** C37H51N7O9S
  • **Molecular weight:** 813.9 g/mol
  • **Origin:** Synthetic fragment of ACTH (residues 4-10)
  • Storage Considerations

  • Store lyophilised Semax at -20°C.
  • Protect from light and moisture.
  • Reconstituted solutions should be aliquoted and stored at -20°C.
  • Testing Methods

Semax batches should be verified by:

  • RP-HPLC for purity (≥98% recommended)
  • Mass spectrometry for identity confirmation
  • Research Limitations

Semax is supplied for in vitro laboratory research and animal model studies only. Most published research originates from Russian research groups. No clinical trials outside Russia have confirmed efficacy. No medical, therapeutic, or dosing claims are made. Semax is not approved for human therapeutic use by Western regulatory authorities.
